The checklist includes the following points:<br /><br />- Review patient history, including age, sex, and test indication.<br />- Perform both chromosome analysis and FISH/microarray studies.<br />- Prioritize FISH for low volume or insufficient volume specimens.<br />- Consult with ordering physician/pathologist for inadequate samples and recommend optimal testing based on guidelines.<br />- Isolate/enrich plasma cells for FISH/microarray testing.<br />- Add appropriate mitogens, if necessary, to the cultures.<br />- Analyze 20 cells when possible and from two or more cultures.<br />- Do not cherry pick cells for analysis, analyze poor quality cells as they may represent the clone.<br />- Prepare an appropriate number of karyograms.<br />- Perform FISH/microarray testing for 17p13 deletion, t(4;14), t(14;16), t(11;14), t(14;20), chromosome 13 deletions, chromosome 1 abnormalities, and hyperdiploidy.<br />- Establish a database and normal cut-off values for interphase analysis for plasma cell FISH.<br />- Include risk assessment in the final report.<br /><br />This checklist serves as a guide for healthcare providers to ensure thorough testing and analysis of plasma cell disorders and to provide a comprehensive assessment of the patient's condition.
